Oversaw and managed the analytical services and training at the Complex Carbohydrate Research Center, which includes three resource centers, Department of Energy (DOE) Resource Center for Plant and Microbial, National Institutes of Health Center for Glycotechnology and National Institutes of Health for Glycomics. The service program enables polysaccharides, therapeutic glycoproteins and glycolipids to be analyzed as a fee-for-service bases for the pharmaceutical, biotechnology and research facilities. Have worked closely with a number of pharmaceutical companies on developing their carbohydrate based drugs, and their needs for out-sourcing prior to phase I and phase II clinical trials. Structural characterization of biologically active molecules such as plant and microbial polysaccharides and lipopolysaccharide and glycolipids and glycoproteins have been carried out by mass spectrometry (MALDI-MS, ESI-MS and ESI-MS/MS), HPAEC, GC-MS and NMR. The responsibilities included: management of each research and service project in the laboratory, writing contracts and agreements for each project, budgeting and invoicing in addition to interactions with clients at all levels; coordination of all three resource centers within the service program; allocating the appropriate resources and personnel to each project and overseeing that each project is completed in a timely manner; making presentations to clients on the scientific data and the direction of each project. The job involved multi-tasking and being able to simultaneously manage more than twenty short and long term projects being carried out. Hiring and training has been under the direction of this consultant. This consultant has also been responsible for all quality assurance aspect of projects in particular standardizing methods for GLP/GMP studies.

Routinely used analytical techniques such as LC-MS, MALDI-MS, FAB-MS, HPAEC-PAD and GC-MS for analysis of carbohydrates Performed Linkage and composition analysis on all glycoconjugates. Organized and taught courses in mass spectrometry and carbohydrate chemistry. Trained staff on use of the LC-MS, MALDI-MS and the FAB-MS instruments. Collaborated with scientists outside the center for structural elucidation of glycoproteins and identification of ‘O’ and ‘N’- linked oligosaccharides using mass spectrometry.

Structural characterization of pectin polysaccharide rhamnogalacturonan I, (RGI) Studied the effect of the enzymes endohyrolase and endolyase on rhamnogalacturonan I, and characterization of the fragments produced by these enzymes by ESI-MS and ESIMS/MS. During this time, I also performed analytical service work for the DOE Center for Plant and Microbial Complex Carbohydrates at the center. The analytical work involved mass spectrometric (FAB-MS, MALDI-MS, LC-MS) analyses of plant- and microbial-derived carbohydrates and composition and linkage analysis. Trained staff on use of the LC-MS, MALDI-MS and the FAB-MS instruments.

Study Director/Senior Scientist, M-SCAN LTD (Analytical Mass Spectrometry

Responsible for complete structural characterization of native and recombinant proteins and glycoproteins as a service to the pharmaceutical industry using enzymatic digestion, HPLC and mass spectrometry (LC-MS and FAB-MS). Designed experiments for analyzing S-S bridged peptides by LC-MS. Developed new mapping experiments using HPLC/FAB-MS for identification of O- and N-linked glycopeptides from a mixture. Performed composition and linkage analysis on oligosaccharides. Prepared all biochemical scientific operating procedures and reviewed reports, which were often submitted, to the FDA. Supervised all quality assurance aspects of each project carried out under GLP.
